| Summary: | This work purpose is to establish protocol for vegetal tissue culture (important tool to preserve germoplasm and to maintain endangered species) with the species Blutaparon portulacoides (A. St.-Hil.) Mears, this species occurs between the upper high tide limit and the dunes and is endangered by the anthropic action. Therefore, the biolgical essay, biochemical procedures and vegetal biotechnology methodology adequacy will be proceeded. The vegetal material was collected in its natural habitat on the coastal area in two locations: Maricá (RJ) and Ubatuba (SP); after that, it was grown in a greenhouse, thus being a vegetal material source for the whole work. Explants (stem segments and most apical portions) were collected and submitted to in vitro experiments, they were distributed in 6 M&S culture means variations (MURASHIGE; SKOOG, 1962), for each variation had 30 repetitions; the piliferous zone development and the average length of main roots were assessed. With the treatment with 40g/L Sucrose and half of M&S standard means macronutrients was present among all groups that presented the best result within the experiment, being an alternative to this species micropropagation. |
